The Mononobe and the Soga were actually two rival clans in Shoutoku-era Japan. The character Tojiko is supposedly based off of was a consort for Prince Shoutoku. Futo doesn't appear to be based off of anyone in particular.

The characters probably knew each other before they all tried to go full shikaisen, but it's very unlikely they're related by blood.
